Expression and significance of HER2 and PTEN in human gastric carcinoma tissue

Abstract

Objective To evaluate the expression and significance of human epidermal growth factor receptor 2(HER2) and phosphatase and tensin homolog deleted on chromosome ten(PTEN) in human gastric carcinoma tissues.Methods The expression levels of HER2 and PTEN protein in 407 human gastric carcinoma tissues were measured by EnVision immunohistochemistry method in tissues-microarry.The results were compared to those in 30 samples of normal gastric mucosa tissues.The relationships between their expressions and clinicopathological features were analyzed.ResultsPositive expression rate of HER2 was 9.1%.Positive expression rate of PTEN was 20.1%,which was significantly lower than 100% in normal gastric mucosa(P0.01).The expression of HER2 was significantly correlated with differentiation and Lauren′s classification(P0.05),which of PTEN was significantly correlated with differentiation,Lauren′s classification and lymphnode metastasis of gastric carcinoma(P0.05).The expression of HER2 was negatively correlated with that of PTEN(r=-0.344,P0.01).Conclusion The expression of PTEN is closely related with occurrence and development of gastric carcinoma.Combined detection of HER2 and PTEN proteins may benefit to evaluate the malignancy and prognosis of gastric cancer.
