Abstract
Firstly,single factor experiment method was used to find the range of each influencing factor.Then,the extraction process of polysaccharides from Ginkgo biloba Leaves was optimized with Uniform Design.The best extraction process for crude polysaccharides was listed as follows:extraction time was 210 min,liquid-solid ratio was 30∶1(mL/g),extraction temperature was 80℃,and extraction times was twice.Under the optimal extraction condition,the highest extraction rate obtained was 7.14%.Verification results was very close to the theoretical value 6.96%.And then the phenol-sulfuric acid method was used to determine the polysaccharide content of each experiment and polysaccharide yield was chosen as indicators for further optimization of the extraction process too.Finally the best extraction conditions for pure polysaccharide was listed as follows:liqiud-solid ratio 35∶1,extraction time210 min,temperature 100℃,the verification results shows that the real yield and the theoretical values were also very close.It proved the process parameters obtained by Uniform Design was reliable.
