Simultaneous Determination of Oleic Acid and Linoleic Acid in Rat Plasma by GC

Abstract

Objective:To develop and validate a gas chromatography(GC) method for the simultaneous determination of oleic acid and linoleic acid in rat plasma.Methods:Chromatographic separation was performed on OV-17 capillary column(30m×0.32mm×0.50μm) with a temperature of 220℃,and a flame ionization detector.Results:The GC method had shown good chromatographic resolution for Methyl oleate and Methyl linoleate.Linearity was tested ranging from 23.80 μg·mL-1 to 2380 μg·mL-1(r=0.9996) and 24.51μg·mL-1 to 2451 μg·mL-1(r=0.9997).The extraction recoveries for two components were between 75.83% and 84.17% and the method recoveries were between 95.85% and 107.1%.The intra-and inter-day RSD were less than 4.7%.Conclusion:The proposed method is simple,accurate and suitable for the determination of oleic acid and linoleic acid in rat plasma and further pharmacokinetics study.
