Abstract
Objective To establish a method for the determination of 7 free fatty acids in human serum.Methods By adding n-heptadecanoic acid as internal standard,stearic acid,oleic acid,linoleic acid,Linolenic acid,arachidonic acid,eicosapentaenoic acid and docosahexaenoic acid in human serum were methylesterified by H2SO4/methanol,extracted and concentrated by n-hexane,detected by gas chromatography.Results After methyl esterification,7 kinds of fatty methyl ester could be separated completely,with a good reproducibility.The linearity was ranged from 5.0μg/ml~500μg/ml(r0.999),minimum limited determination was ≤1.1μg/ml.The average recovery rate of 3 levels(20μg/ml,100μg/ml,300μg/ml) was between 91.7% and 98.1%,RSD was from 1.9% to 4.1%.Conclusions The method can be used to the determination of free fatty acids in human serum.
