Structure and Sequence Analysis of 18s rDNA and ITS Gene of Two Yeast Strains Producing Ginsenoside Glycosidases

Abstract

The sequences of 18S rDNA and ITS gene fragments of two yeast strains which can produce ginsenoside glycosidases are cloned and analyzed,and both 18S rDNA and ITS gene sequences are obtained.It is shown from the homologous analysis results that the sequence similarity of 18S rDNA and ITS fragments from two yeast strains is 100% and 66% respectively,and the 18S rDNA and ITS gene sequences are highly homologous to the relevant sequences of Saccharomyces cerevisiae in GenBank.The ITS sequences of 10 yeast species which belong to different genera are retrieved from GenBank,and the phylogenetic tree is then constructed by using these sequences and neighbor-joining method.The results from molecular biological analysis show that these two yeast strains are identified as different species of Saccharomyces.
