Sequence Analysis on 5.8 S rDNA and ITS of Yeast from Koumiss

Abstract

The main purpose of the experiment was to extract genome DNA from the two yeast strains that were isolated from koumiss,and to amplify 5.85 rRNA and ITS gene by use of a pair of primers,namely,ITS and ITS4,then connect with the pRNA vector.Agter cloning,the sequence determination was conducted.Scientists made homologous analysis on the obtained sequence and the sequence of GenBank data base and system evolutionary tree was established.Combined with analysis of the phylogenetic tree and partial sequences of 5.8S rDNA and ITS,J14 and S33 was classified as Kluyveromyces marxianus.The innovation stress in this experiment is that:the traditional yeast taxology in koumiss mainly depends on morphological,physio-biochemistry and other identification methods.In this experiment the 5.85 rRNA and ITS genes were firstly determined with molecular biological method.
