Analysis on Genetic Diversity of Xinjiang Pears Based on the SRAP Marker

Abstract

This study was carried out to reveal the genetic diversity of 40cultivars of Xinjiang pears by use of molecular marker technique SRAP(sequence-related amplified polymorphism).The forward primers and revorse primers were composed and amplified to be screened,among which 10pairs of primers with strong specificity and stability were analyzed by SRAP.Altogether 131bands were amplified from the 10 pairs of primers,among which there were 127bands with polymorphic loci.The total of 13.1bands with polymorphism were amplified from each group of primers.At species level,the average percentage of polymorphic bands in all primers was 96.95%.SRAP genetic relationship dendrogram Cluster analysis showed that 40pear germplasm based on the 0.625genetic similarity coefficient is divided into three categories:first category group included 2germplasm;the second category included 6germplasm;the third category included 32germplasm.
