Abstract

OBJECTIVE To purify the total flavonoids fromPolygonum orientle L with macroporous adsorptive resin and establish optimum process for separating and purifying total flavonoids.METHODS The optimal techniques of separation and purification of flavonoid were obtained by static adsorption and elution tests,and separation techniques were evaluated by measuring the concentration of total flavonoids fromPolygonum orientle L with ultraviolet spectrophotometer.RESULTS The type D101 macroporous adsorptive resin was the best for the separation and purification of total flavonoids of Polygonum orientle L.The optimum process condition was sample amount 103.3 mg·g-1,and sample concentration2.213 mg·mL-1 at pH4;and eluting reagent was 50% alcohol with 4 BV,and water elution volume was 3 BV.The recovery rate of total flavonoids reached56.2% with the purity of 72.3%.CONCLUSION To utilize the D101 macroporous adsorptive resin for the separation and purification of the total flavonoids Polygonum orientle L is feasible,shows high recovery rate and purity.
