Genetic Relationship Analysis between Wild Species and Cultivars of Rubus L.by ISSR Marker

Abstract

Genetic relationship of 39 Rubus L.wild species or varieties and 17 cultivars were studied using inter-simple sequence repeat(ISSR) marker.12 primers,which were screened out from 35 primers,could get clear and polymorphic fragments.A total of 166 DNA fragments ranging from 259 bp to 2300 bp were got through PCR by these 12 primers.The average number of DNA fragments produced by each primer was 13.83.The polymorphic rate was 96.33 %.A dendrogram of showing genetic relationships was constructed by using the NTSYS-pc software and UPGMA.The 39 wild species and 15 cultivars were divided into 5 groups by clustering analysis,and other 2 cultivars(Kotata and Boysen) belonged to another groups and differented from the 54 a materials.The results showed that ISSR molecular marker technique was suitable for analyzing the Rubus interspecific relationship.
