Abstract
The protein hydrolyte was prepared by hydrolysis tilapia fishbone with neutral protease.Taken hydrolysis degree as an optimum index,the effect of initial pH,hydrolysis temperature,solid-liquid ratio,and enzyme dosage and hydrolysis time on hydrolysis degree was studied.Based on single factor experiments,the optimum process condition was obtained by using Box-Behnken center-united design and surface response design.Under this condition,the effect of different pre-treatment methods including water bath,high pressure steam,ultrasonic wave,and microwave on hydrolysis degree were studied.The result showed that the optimum process condition was temperature 49℃,enzyme dosage 1 824U/g,hydrolysis time 6h and the hydrolysis degree of tilapia fishbone 33.7%.Most pre-treatment methods were harmful to subsequent hydrolysis of tilapia fishbone,expect 110℃ high pressure steam and ultrasound with the power of 180W.
