Expression of Osteopontin in Rat Peritoneal Mesothelial Cells Stimulated by High Glucose and Lipopolysaccharide

Abstract

Objective To observe the osteopontin(OPN) expression in rat peritoneal mesothelial cell(RPMC) stimulated by high glucose and lipopolysaccharide.Methods RPMCs were isolated from rat colicomentum and incubated with high glucose and lipopolysaccharide(8,12,24,34,48 h).The RPMCs were stimulated with different concentrations of high glucose(1.5%,2.5%,4.25%) and lipopolysaccharide(50,100 μg/mL).RPMC in the control group were only incubated with regular medium.OPN and TGF-β1 was detected by immunohistochemistry and/or real-time RT-PCR.Results Compared with the control group,the expression of OPN was significantly increased in the cells stimulated by lipopolysaccharide and high glucose.Compared with the group stimulated by lipopolysaccharide and glucose,the up-regulating expression of OPN and TGF-β1 in cells pre-incubated with lipopolysaccharide and high glucose was significantly inhibited.Conclusion LPS and high glucose can up-regulate the expression of OPN and TGF-β1 in RPMC.
