Abstract
Objective:To observe the acute toxicity of Jubaoluyou Capsule(JC) in mice and its long-term toxicity in rats.Methods:To determine the maximum tolerance dose(MTD) of the new agent, the mice were given the maximum concentration and the maximum volume of JC by ig administration.80 SD rats were randomly divided into four groups:high dose group, medium dose group, low dose group and control group.JC was continuously administrated for 90 days. When the test was finished, rats were killed and the following indexes were determined:growth and development status,blood cell counting and some biochemical indexes. Histopathological examinations were simultaneously performed. After withdrawal 15 days, the above-mentioned indexes were detected.Results:The MTD of JC by ig administration in mice was greater than or equal to 21.6 g/kg. In the long-term toxicity test, the kidney in the high-dose group had the following pathological changes:the epithelial cell of proximal convoluted tubules slightly swelling in two, minor blood vessels in renal medulla becoming bloodshot and interstitial lymph-cell infiltration in two.After withdrawal 15 days, these pathological changes could be completely recovered. The other indexes had no significant difference between the three treated groups and the control group.Conclusion:JC administration by long-term might slightly injure the renal function, but the injury was reversible. It is safe to administrate the prescribed dose of JC.
