Effect of antisense c-myc oligonucleotide on telomerase subunits in SMMC-7721 hepatocarcinoma cells

Abstract

Objective:To investigate the anti-cancer mechanism of antisense c-myc furthˉery,the effect of antisense c-myc on telomerase subunits and c-myc protein of SMMC-7721hepatocarcinoma cells was observed.Methods:Antisense c-myc was delivered to cultured SMMC-7721.Using immunohistochemistry、RT-PCR,the change of c-myc protein exˉpression and telomerase-related major subunits hTR、hTERT、TP1mRNA was observed.Results:Antisense c-myc,at concentration of5000U/mL,can successfully inhibit the exˉpression of c-myc protein and hTERT mRNA in SMMC-7721hepatocarcinoma cells(P0.05or P0.01),but can not inhibit the expression of TP1and hTR mRNA(P0.05).Conclusion:The data suggested that antisense c-myc can inhibit the expression of telomerase hTERT and there is no relation to the expression of hTR and TP1,which could be the major mechanism of antisense c-myc inhibiting telomerase activity.
