Abstract
Objective Enamel matrix proteins(EMPs) can promote the regeneration of periodontal tissue.Simvastatin can enhance cell ossification activity,reduce the incidence of fracture,and prevent osteoporosis.The aim of this study was to observe the effects of the combination of EMPs and simvastatin on proliferation and alkaline phosphatase(ALP) activity of bone marrow stromal cells(BMSCs).Methods Acetic acid method was used to separate and purify the EMPs of pig,and western blot was used to detect the EMPs.Whole bone marrow method was used to culture rat BMSCs,and BMSCs were treated with different concentration of EMPs and simvastatin.MTT method was used to examine the cell proliferation,enzyme kinetics method was used to examine the ALP activity of BMSCs.The biological character of BMSCs was observed after the treatment with the best concentration of the two drugs.ResultsBoth the proliferation and ALP activity of BMSCs were increased after the treatment with EMPs and simvastatin.Compared with the separate application,there was no differences in proliferation but had significant difference in ALP activity after the combined application.Conclusion The combination of EMPs and simvastatin could enhance the differentiation of BMSCs.
