Abstract

The surface disinfactant and sterilized time,active carbon,VC in use,dark treatment time,light intensity and interval time for transferring another bottle were studied to reduce the browning rate of explants in tissue culture of G.conopsea.The results showed that the browning rate could reduce from 93.33% to 48.28% when the explants were sterilized by 0.1% mercury solution for 10 min,the explants were transferred into another bottle every three days and the treated explants were cultured on MS + KT 0.5 mg/L +ZT 0.5 mg/L+NAA 1.0 mg/L+AC 1.0 g/L+VC 2.0 mg/L without light at the early time of tissue culture for 5~10 d,light intensity 2 000 lx was the best.
