Chemosensitivity tests of oral cancer by MTT assay. Studies on fresh tumor cells.

Fresh surgical specimens from 14 oral cancer patients were used for chemosensitivity tests by MTT assay.The chemosensitivity of 11 out of 19 samples taken from the patients was able to be assessed. The mean optical density (OD) of the control culture containing no drugs was 0.163, which was suitable for measuring the fine sensitivity of cells against various anti-cancer drugs. Squamous cell carcinomas (SCC) were highly sensitive to cisplatin (CDDP), followed by peplomycin (PEP) and 5-fluorouracil (5-FU).Synergistic growth inhibitory effect was detected against 2 SCC when PEP was used in combination with small amounts of CDDP. Thus, MTT assay was considered to be a useful chemosensitivity screening test for oral cancers.
