In Vitro Callus Induction and Plantlet Regeneration Studies through Anther Culture in Two Indica Rice (Oryza Sativa L.) Varieties

Abstract

Effect of organic adjuvants, synthetic plant growth regulators and diverse carbon sources in influencing anther culture response in two indica rice genotypes (GR 11 and Gurjari) were assessed due to genotypic difference. Androgenic callus induction as well as green plantlet regeneration was more when callus was induced on N6 fortified with 100 mgl−1 yeast extract (YE). However, casein hydrolysate (CH) at 50, 250 and 500 mgl−1 concentrations encouraged green plantlet regeneration. High callus induction was observed with synthetic growth regulators in comparison with the control set. However, green plantlet regeneration was observed when callus induction medium (CIM) was supplemented with 2, 4-D, NAA and Kn in both the varieties. Between two varieties, Gurjari showed better callus induction on N6 medium supplemented with 6% sucrose. Maximum callus induction was observed in Gurjari when 6% sucrose was used as carbon source. Plantlet regeneration in the present study was found to be very low. This study elucidates that the genotypic differences to in vitro response and the probability of the exploitation of androclonal variation in indica rice.
