TTF-1 gene cloning and DNA polymorphisms analysis in sheep

Abstract

In this study,two coding fragments of TTF-1 gene were amplified and cloned with PCR.Then sequences were submitted to Blastn searching in NCBI,and accepted by GenBank(Accession No:DQ010919 and DQ010920). Homogeneous analysis demonstrated that nucleotides and amino acids sequences of sheep TTF-1 gene between different species mammals were comparatively conservative.Sequences analysis showed that two fragments contained 318 and 249 bp in length and encode two polypeptides containing 105 and 82 amino acids,respectively.In addition,PCR-SSCP results showed that there were no polymorphism in nucleotide sequences of TTF-1 gene among three sheep breeds of different litter size.
