Molecular cloning and sequencing analysis of interleukin-2 gene from 10 kinds of Guangxi local chickens

Abstract

A pair of specific primer was designed according to the sequences of chiken interleukin-2 gene(ChIL-2) published in GenBank. The specific 470 bp fragment of ChIL-2 products was amplified by Reverse Transcription-Polymerase Chain Reaction(RT-PCR) from ten kinds of Guangxi local chickens' spleen lymphocytes which were activated by concanavalin A(ConA).After the RT-PCR products were cloned into pMD18-T vetor,the nucleotide acid sequences were identified by PCR and restrictive endonucleases digestion.The results showed that the coding region of genes encoded a predicted mature protein of 143 amino acids with a molecular weight at 16.3 ku.The sequence homology was 24.8%~30.3% and 98.8%~99.8% in nucleotide acids,14.6%~16.7% and 96.5%~98.6% in amino acids,compared with mammals and others chickens.
