Effect of kinetin and IAA on micropropagation of orchid lily (Barclaya longifolia).

Abstract

Orchid lily Barclaya longfolia in the wild were considered to be decline and slow propagation by seed or rhizome. Micropropagation technology to help solve this problem. The sterile tissue was cultured using the combination of 6-furfurylaminopurine (kinetin) at 0, 1, 2, 3 mg/L and 3-indoleacetic acid (IAA) at 0.0, 0.5, 1.0, 1.5 mg/L supplemented in MS (Murashige and Skoog, 1962) liquid medium. After 8 weeks, it was found that the MS liquid medium containing kinetin 2 mg/L and IAA 1.0 mg/L increased the number of plantlet and new leaf significantly (P LT 0.05). The average numbers of plantlet and new leaf were 2.52+-0.56 plantlet and 33.50+-4.09 leaves, respectively. Adding a large amount of IAA tended to decrease the number of plantlet in B. longfolia in vitro culture. Consequently, our results further demonstrate that the micropropagation of B. longfolia provides an efficient method to rapidly produce salable quality aquarium plants without the need for field collection.
