Effects of different factors on the germination and growth of Pyrrosia lingua spores.

Abstract

The mature spores of Pyrrosia lingua were used as explants,the effects of different sugar concentrations,growth regulators,and culture media on the germination and growth of the spores were investigated.The results indicated that adding 1%-2% sugar in the culture medium was beneficial to the germination of spore,and the germination rate of spore was over 80%.The culture medium with low salt such as 1 / 8 MS and Knop,s could also promote the germination of spore,and the germinating stage was six days and the germination rate was 83.2% and 84.1%,respectively.Adding 1.0 mg / L NAA or 0.5 mg / L 6-BA to the culture medium could shorten the germination stage of spore,and had the highest germination rate.Moreover,1 / 2 MS culture medium was more suitable for the propagation of prothallus,and the incomplete culture medium with the river sand was more suitable for the induction of sporophyte and the induction rate was over 90%.
