Abstract

In vitro experiments were conducted to test the effects of culture medium types, content of sucrose, light intensities, temperatures and Pb~(2+)concentration on spore germination of Polytrichum commune(Polytrichaceae,Musci). Results show:the culture medium with rich nutrient promotes the bipolar germination rate of the spores, while those with low nutrient increase the spore germination rate.Whether light is present or absent, the spore germination rates are similar on the media with minerals. However, the light significantly inhibits the germination of the spores on SM medium without minerals. In the media with 0.5%sucrose under the temperatures from 20~30℃,the spores germinated well. With the increase of the Pb~(2+)concentration in the medium, the spore germination rate decreases significantly. The response of the spores to the factors in virto are speculated from their habitat characteristics in the field. Our results preliminarily reveal the factors to facilitate or inhibit the spore germination, which is valuable for us to culture P. commune both in the field and in vitro conditions.
