Abstract
For polysaccharide production, both the one-factor-at-a-time and SAS methods were applied to the optimization of submerged fermentation by Inonotus obliquus. The best components of the medium were determined as following:corn syrup 2.11%,soluble starch 2.23%,yeast powder 0.51%,peptone 0.41%,KH2PO4 0.2%,and magnesium sulfate 0.05%.Using the optimum media, the culture pH,reducing sugar, polysaccharide and mycelia biomass of Inonotus obliquus in 12 days were discussed. Results showed that, during the fermentation, constant pH value was found. The mycelia biomasses was increased firstly and then reached the maximum value of 1.35 g/L at 6 days, while the crude polysaccharide content was increased firstly and then decreased later with its highest value of 3.85 at 7days.
