Expressions of cyclooxygenase-2 mRNA and heparanase mRNA in renal cell carcinoma.

Abstract
Objective To detect mRNA expressions of cyclooxygenase-2(COX-2) and heparanase(Hpa) in renal cell carcinoma(RCC),and to investigate their correlation with carcinogenesis,development and prognosis of RCC.Methods Semi-quantitative reversed-transcription polymerase chain reaction(semiquantitative RT-PCR) was used to detect mRNA expressions of COX-2 and Hpa in 56 samples of RCC tissues and 28 samples of the adjacent-tumor renal tissues,and β-actin served as a quantitative standard marker.Different expressions in RCC tissues and adjacent-tumor renal tissues were compared and their relations with clinical pathology parameters and prognosis were analyzed.Results Among 56 cases of RCC tissues,the positive rate of COX-2 mRNA and Hpa mRNA expressions was 73.2% and 67.9%,both were significantly higher than those in the adjacent-tumor renal tissues (χ2=17.719,P0.05;χ2=21.429,P0.05).Low grade(Fuhrman grades 1 and 2) RCC showed a higher COX-2 mRNA expression than high grade ones(Fuhrman grades 3 and 4)(χ2=30.727,P0.05).The relationship between COX-2 mRNA expression and prognosis of RCC patients was not significant(χ2=0.890,P0.05).Expression of Hpa m RNA was related to Fuhrman grade of RCC(χ2=7.718,P0.05),clinical stage(χ2=3.963,P0.05),and lymphatic metastasis(χ2=3.852,P0.05).The relationship between Hpa mRNA expression and prognosis of RCC patients was significant(χ2=12.72,P0.05).There was no relationship between COX-2 mRNA and Hpa mRNA expression(r=-0.244,P0.05).Conclusion COX-2 mRNA expression is an early molecule occurrence of RCC genesis.Hpa mRNA expression can predict the invasion and metastasis of RCC and can be used as a marker for prognosis of RCC.
