Abstract

The protoplast preparation and regeneration conditions in Trichoderma reesei 40359 were studied.The results showed that the optimal condition for protoplast preparation in Trichoderma reesei 40359 was culture mycelia for 12 h,then enzymelysed for 2 h at 30 ℃ by the mixture solution of snail digestase(2%) and cellulase(2%).The protoplasts formation number was 6.14×105·mL-1 and the regeneration rate was 10.6% under the optimal condition when 0.6 mol·L-1 surose used as the osmotic stabilizer.
