Abstract
Objective: To study the effect of tryptanthrin(Try) on proliferation and apoptosis of breast cancer MCF-7 cells.Methods: The proliferative effect of Try (1.56-100 μmol/L) on MCF-7 cells was assessed by MTT assay.The morphologic change was observed by transmission electron microscope.The flow cytometer was used to detect the changes of cell apoptosis,cell cycle and mitochondria membrane potential.Results: MTT results showed that in the range of 12.5-100 μmol/L,Try obviously inhibited the proliferation of MCF-7 cells in a dose and time-dependent manner.Typical apoptosis changes were observed in MCF-7 cells treated with Try for 48 h by transmission electron microscope.To stain with Annexin V-FITC and PI,Flow cytometer results showed that the apoptosis state was obvious in the cells treated with 50,100 μmol/L Try for 48 h,respectively.Try can change the cell cycle distribution of MCF-7 cells.The G1 phase was blocked and the DNA synthesis was inhibited.Meanwhile,the mitochondrial membrane potential was decreased.Conclusions: Try has an effect on inhibiting the cell proliferation and inducing the apoptosis of MCF-7 cells.
