Abstract

Objective To study the inhibitive effect of transfected PTEN on LoVo cells. Method The human PTEN cDNA was transfered into LoVo cells via lipofectin and PTEN mRNA levels and its expression were analysized by flow cytometry. Before or after transfection, the activities of 5-Fu in inhibiting cell proliferation and inducing apoptosis were measured by flow cytometry and MTT. Results PTEN transfection significantly up-regulated PTEN expression in LoVo cells. 5-Fu inhibited cell proliferation and induced apoptosis in transfected LoVo cell. Conclusion Transfected PTEN remarkably up-regulated PTEN expression in LoVo cells and promoted the apoptosis. PTEN transfection associated with 5-Fu treatment had cooperatively cytotoxic effect.
