Abstract
【Objective】In vitro culture of Melaleuca viridiflora ‘Red' was studied in order to provide technique support on its industrialized seedling production. 【Method】Stem segment with one axillary bud was used as explant to investigate the effects of different hormone combinations on adventitious bud induction, cluster bud multiplication and rooting induction. 【Result】The axillary bud of stem segment could be induced into adventitious bud and cluster buds in multiplication culture medium. The multiplication rate of buds in MS medium adding with different combinations 6-BA 0.1-2.0 mg/L +NAA 0.01 mg/L and 6-BA 0.1-1.0 mg/L +IBA 0.01 mg/L showed increasing firstly and then decreasing tenderly. MS+6-BA 0.5 mg/L+ IBA 0.01 mg/L was the optimizing culture medium of bud multiplication, and the multiplication rate was 2.2times. Adding 0.1-0.5 mg/L NAA or IBA in 1/2MS medium, the rooting rate of buds increased significantly, which reached 100.0% or 56.0%-90.0%, and the average root number was 12.6-14.8 and 3.1-4.8, respectively. The suitable rooting medium was 1/2MS+NAA 0.1-0.3 mg/L. AC 300 mg/L was not suitable for bud multiplication and rooting induction. The survival rate of transplanting test-tube plantlets reached 99.0% using peat as culture medium in conventional management. 【Conclusion】The regeneration system of Melaleuca viridiflora ‘Red' was established, and stem in vitro culture was an effective path for rapid propagation.
