CYTOPATHOLOGICAL OBSERVATION OF RICE STRIPE

Abstract

By electron microscope and immunogold labeling technique, the cytopathological alteration of rice stripe virus infected leaves, and the distribution of the virus particles, the coat protein and the disease specific protein in the cells were observed. In mesophyll cell, the quantity of the mitochondria increased and the nuclear enlarged. There were protein bodies in most of mesophyll cells. The ultrastructure of chloroplast granum of the disease leaves was altered. Its granum appeared sparse and thick, or was forced tightly and became thin. In some cases the chloroplast was destroyed, while the rested granum remained in the cytoplasm. In the mesophyll and sieves in vascular bundle of the disease leaves, there were some inclusions and bodies which showed higher electronic density. With higher amplification they were dots and filaments about 8 nm in diameter. The coat proteins and the disease specific protein were found in the chloroplast, cytoplasm and nuclear, but not in mitochondria.
