CLINICAL STUDY OF NEDAPLATIN COMBINED WITH CISPLATIN FOR THE TREATMENT OF 26 PATIENTS WITH ADVANCED ESOPHAGEAL CARCINOMA

[Objective] To evaluate the efficacy and adverse events of diminished dose of nedaplatin(NDP)combined with low dose of cisplatin(DDP)plus 5-Fluorouracil(5-Fu)/Folic Acid in the treatment of advanced esophageal carcinoma.[Methods] The patients were given NDP(25 mg /m2,iv gtt)and(DDP 15 mg /m2,iv gtt)on day 1 and day 8 and continu-ously infused with 5-Fu(300 mg /m2)for 24 h on days 1-5 and days 8-12.Folic acid tablets(60 mg /d)were given to the pa-tients following infusion of 5-Fu.The therapeutic regimens were repeated every 21~28 days(one cycle).The efficacy was eval-uated after two cycles of treatment.[Results] The median cycle of treatment was 4.5.The overall response rate was 61.5%(16 /26),including 2 cases of CR,14 cases of PR,5 cases of SD and 5 cases of PD,respectively.The major adverse event was bone marrow suppression.There was no grade IV bone marrow suppression.Non-hematologic toxicities including nausea,vomiting,stomatitis,liver and renal function impairment were less toxic.[Conclusion] Diminished dose of NDP combined with low dose of DDP plus 5-Fluorouracil /Folic Acid is effective and safe in treating patients with advanced esophageal carcino-ma.
