Pharmacodynamics and dosage of propofol in two concentration during anesthesia of patients with laparoscopic dholecystectomy

Abstract

Objective To compare the pharmacodynamics and dosage of propofol in different concentration during anesthesia of patients with laparoscopic cholecystectomy. Methods One hundred patients with laparoscopic cholecystectomy were randomly divided into two groups (n=50). Group Ⅰ was given 1% propofol (2 mg/kg, iv) and group Ⅱ was given 2% propofol (2 mg/kg, iv) for anesthesia induction. Depth of anesthesia was monitored by Narcotrend anesthesia depth monitor during the whole procedures. Infusion speed was adjusted through Narcotrend index. The time of consciousness extinction, Narcotrend index value drops to 36, Narcotrend index value recuperates to 65 and the dosage of propofol in first hour were observed and recorded. The drop percentage of MAP and HR within 15 min after induction starts were also assessed. Results The time of consciousness extinction, and Narcotrend index value drops to 36 were (115±45) s and (136±54) s respectively in group Ⅰ, and were (156±60) s and (183±61) s respectively in groupⅡ. The values in groupⅠare significantly shorter than values in GroupⅡ(P<0.05). The total dosage and the dosage of propofol in first hour of groupⅠwere (41±15) ml and (36±10) ml respectively. The double volume of total dosage and double volume dosage of propofol in first hour were (53±18) ml and (46±15) ml respectively in groupⅡ. The double dosage volume of groupⅡis more than total dosage volume of groupⅠ(P<0.05). Conclusions The potency of 1% propofol is stronger than 2% propofol. Key words: Propofol; Pharmacodynamics
