Abstract
Objective To approach the effects of size-separated titanium particles and the ratio of titanium particles to osteoblasts on low-density lipoprotein receptor 5 ( Lrp5 ) mRNA in osteoblasts.Methods (0.35 ± 0.09) μm titanium particles were co-cultured with osteoblasts in vitro at various particle number to osteoblasts ratios ( 1∶ 1,10∶ 1,100∶ 1,500∶1 ) and ( 1.5 ±0.38) μm,(6.25 ±2.13) μm titanium particles at rate 10 ∶ 1 with osteoblasts.The co-culture supernatants were analyzed at the mRNA level by using reverse transcriptase polymerase chain reaction (RT-PCR).Results Weard titanium particles significantly downregulated Lrp5 mRNA expression in osteoblasts.Compared to the normal control group,the expression of Lrp5 mRNA was significantly reduced in osteoblasts in the other groups (P < 0.05 ).Osteoblasts co-cultured with (0.35 ±0.09) μm titanium particles,compared to those co-cultured with ( 1.5 ±0.38) μm and (6.25 ±2.13) μm titanium particles,significantly restrained Lrp5 mRNA expression (P <O.05 ).Conclusion The size and concentration of titanium particles are important parameters to decide the cytobiologic model of osteoblasts.Smaller size and higher concentration of weared titanium particles can more significantly inhibitied the Lrp5 mRNA expression.
