Research on the change trend of key molecules in PI3K/Akt signaling pathway after spinal cord injury

Abstract

Objective To understand the PI3K / Akt signaling pathway molecule trends in time course after spinal cord injury. Methods Animal model of spinal cord injury, divide it into normal group, the group of one day after spinal cord injury, the group of spinal cord injury after 3 days, 7 days after spinal cord injury group, the group of 14 days after spinal cord injury, spinal cord injury group after 28 days . Observed the temporal variation of key molecules in the PI3K/Akt pathway PI3K, Akt, p-Akt after spinal cord injury through immunohistochemistry and Western-blot method. Results Immunohistochemical staining results showed that the distribution of PI3K in spinal cord tissues of normal control group was less. The expression of PI3K was the highest in the 1 days after spinal cord injury, and the expression of was gradually decreased, but the absolute expression level was higher than that in normal control group (P<0.05). The distribution of Akt in normal control group was less than that in the control group, the expression of Akt was the highest in 1 days after spinal cord injury, and the expression of in the normal control group was lower than that in normal control group (P<0.05). Western-Blot showed that the expression of p-Akt/Akt and PI3K was the highest in the 1 days after spinal cord injury, and then decreased gradually in the other group, but the absolute expression level was higher than that in the normal control group (P<0.05). Conclusions PI3K,Akt and P-Akt in the PI3K/Akt signaling pathway increased rapidly after spinal cord injury, and then decreased gradually. It showed that PI3K/Akt signaling pathway was involved in the secondary injury of spinal cord, which may be related to apoptosis. Key words: Spinal cord injury; Apoptosis; PI3K; Akt
