Inhibition of rat cirrhotic fat-storing cell proliferation induced by bone marrow stromal cells

Abstract

Objective To investigate the growth inhibiting role of rat bone marrow stromal cells(BMSCs)on the cirrhotic fat-storing cells(CFSCs).Methods CFSCs and rat liver cell line BRL were cultured with BM SCs culture supernatant collected at different time point.The activities of CFSCs were measured by MTr assay:BMSCs were co-cultured with CFSCs by trans-well insert.The apoptosis of co-cultured CFSCs was detected by terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)assay.Results The supernatant from BMSCs cultures inhibited CFSCs proliferation in a dose-dependent manner,but no inhibition of BRL prolifera tion had been observed.The inhibition of CFSCs proliferation induced by BMSCs was irreversible.The propor tion of apoptotic CFSCs increased significantly as the ratio of BMSCs to CFSCs increaSed.Conclusions BM SCs inhibited CFSCs proliferation in an irreversible manner.BMSCs induced CFSCs apoptosis.
