Abstract
Objective To investigate the effects and mechanisms of resveratrol on proliferation and apoptosis of cervical cancer Hela cells in elderly patients. Methods Hela cells were treated with resveratrol at concentrations of 0 (control group), 50, 100, and 200 μg/ml. The methyl thiazol tetrazolium (MTT) assay was used to test the proliferation and cytotoxicity of resveratrol on Hela cells at 24, 48, and 72 h. The morphological changes of Hoechst33258 cells were observed by fluorescence staining. The expression of B cell lymphoma/leukemia-2 (bcl-2) and bcl-2 associated X protein (bax) was detected by Western blotting. Results Resveratrol 0/50/100/200 (μg/ml) treated with four concentrations of cervical cancer cells, the control group and the experimental group 24, 48, 72 h after cell proliferation was significantly reduced, with the drug concentration increased, the proliferation rate gradually Decreased (P=0.031). The cell proliferation rates at 24 and 48 hours were 0.99/0.87/0.78/0.60, 0.99/0.84/0.67/0.48 respectively. The cell proliferation rates were 0.99/0.83/0.66/0.47 at 72 hours. There was a statistically significant difference (P=0.020). Resveratrol treatment of Hela cells Hoechst33258 staining, Hela cells decreased significantly, the difference between the two groups was statistically significant (P=0.011). Resveratrol could reduce the expression of bcl-2 and increase the expression of bax in cervical cancer cells. And with the increase of drug concentration, the difference became larger (P=0.024). Conclusion Resveratrol can inhibit the proliferation of cervical cancer Hela cells, With resveratrol concentration increased, the proliferation rate was decreased with a concentration-dependent.
