Protective effect of IκBα mutant on the acutep ancreatitis in rats

Abstract

Objective To study the protective effect of the IκBα mutant gene on the acute pancreatitis(AP)in rats.Methods The pancreatitis in rats were induced by cerulean(CR),and the rats were randomly divided into group A(control),group B(AP),group C(AP+PcDNA3.0 transfection),and group D(AP+PcDNA3.0-IκBαM transfection).The expression of TNF-α,Fas,FasL,and Caspase-3 was detected by ELISA and RT-PCR.The necrosis of acinar cells was evaluated by immunohistochemistry.The serum level of amylase in every group was determined.Results In comparison with groups B and C,the expression of TNF-α and the serum level of amylase were decreased significantly,the expression of Fas,FasL,and Caspase-3 mRNA was increaSed signifieiantly,and the number of necrotic acinar cells was,decreased significantly in group D.All pancreatitis groups had significant difference with group A(P<0.05).Conclusion The IκBαM can protect against the acute pancreatitis in rats by inducing the apoptosis of the acinar cells.
