Abstract
Objective To detect the expression pattern of poly adenosine di-phosphate (ADP)-ribose polymerase (PAPR)-1 mRNA and protein in ethanol-induced brain injury in mice and the significance.Methods Ethanol was diluted to a 20% solution in sterile normal saline.Twelve mice of seven-day-old were injected subcutaneously either with ethanol (2.5 g/kg at 0 h and again at 2 h) or with normal saline.The histological changes of brain tissues were observed by HE staining.The expression levels of PARP-1 mRNA and protein were was detected by real-time reverse transcription-polymerase chain reaction (RT-qPCR) and immunohistochemistry (IHC) respectively.Results The relative expression level of PARP-1 mRNA (11.960±2.136,n =6) was enhanced in ethanol group as compared with normal saline solution group (4.333 ± 1.667,n =6) with the difference being significant (P < 0.05).Meanwhile,the yellow-stained substances were seen in ethanol group and the positive expression rate of PARP-1 was 83.3% (5/6).Only little yellow-stained substances were observed in normal saline group and the positive expression rate of PARA-1 was 16.7% (1/6).There was significant difference (P < 0.05).Conclusion The expression levels of PARP-1 mRNA and protein were enhanced in ethanol-induced brain injury model,and the mechanism of PARP-1 activation needs to elucidate in further study.
