An improved method for murine islet isolation and subcapsular kidney transplantation

Abstract

Objective To present a series of islet transplantation in mice by improve the method of islet isolation and subcapsular kidney transplantation.Methods Islet isolation was completed by end of the common bile duct ligation,Perfusion of the pancreas by 3 ml collagenase V (2 g/L),filtration by 100 μm filter and purification by picking islets under microscopy.Islets' yield,purity,function of insulin secretion and effect of islet transplantation were evaluated compared with islets isolated by Ficoll density gradient separation method.Results Purification by filtration increased islet yield (P < 0.01),and resulted in higher insulin secretion in vitro (P < 0.01).Moreover,when syngeneic diabetic hosts were transplanted with a marginal islet mass,the number of islets purified by filtration which restore normoglycemia significantly was less than those isolated by Ficoll.Conclusion Islet filtration is a simple and rapid procedure for purification of islets that demonstrate improved functional mass.The islet transplantation model consisted by Islet filtration and subcapsular kidney transplantation might be widely used in the research of islet transplantation.
