Abstract

Objective This study aimed to improve methods of islet isolation and transplantation in rats.Methods Modified stationary digestion method and islet transplantation using yellow tip method was used in this study.Results ①Time of islet isolation was 164.00±1.74min ②The yield of pureislets was 320.50±44.25/rat ③Purity of islets was 95.90±0 99(%) ④Time of islet transplantation was 27.60±1.71min.⑤At 1~2 days after syngenic islet transplantation,blood glucose level of diabetic rats fell under 200mg/dl.⑥Islets,exocrine tissue and lymph nodes were clearly divided,when ground reflector of microscope was used.Conclusion ①Islet isolation and transplantation method after improvement was more convenient.②Ground reflector of microscope was useful for observation of islets.
