Expression of chemotactic factor macrophage inflammatory protein-2 following rat orthotopic liver transplantation

Abstract

Objective To study the expression of chemotactic factor macrophage inflammatory protein-2(MIP-2)following ischemia-reperfusion injury of liver after rat orthotopic live transplantation. Methods By using rat orthotopic live transplantation model, donor liver was preserved in lactic acid Ringer's solution for 4h (4℃). The rats were then divided into two groups, namely, 1-h after transplantation and 4-h after transplantation. False operation group served as control. The expression of MIP-2 mRNA in liver and protein expression of MIP-2 in serum, neutropil infiltration in liver and ALT of serum were detected. Results In the donor liver pretreated with cold preservation for 4?h, compared with control group, the expression of MIP-2 mRNA and serum albumin in the group of 1-h after transplantation was increased significantly (P<0.01). The expression of MIP-2 mRNA and serum albumin in the group of 4-h was higher than that of 1-h group after transplantation (P<0.01). The expression of MIP-2 mRNA and albumin were positive correlation to neutrophil infiltration and ALT of serum with the correlation coefficient being 0.88, 0.92, 0.90, 0.93 respectively (P<0.01). Conclusion MIP-2 might play an important upregulatory role in the ischemia-reperfusion injury following rat orthotopic liver transplantation and become one of the most important factors in reperfusion injury of liver transplantation.
