Abstract
This study report a method for preparating human sperm chromosome with hamster serum for postinsemination culture of the zona-free hamster ova. The rate of analyzable karyotype was markedly increased. Comparison hamster serum with fetal bovine serum. The average rate to reach the first-cleavage was 102.45% in the inseminated oocytes versus 58.28%. The rate of observable karyotyping in two sera were 76.15% versus 31.51%(P<0.001)。Spontaneous incidence of sperm chromosome aberrations was analyzed in a total of 498 spermatozoa obtained from six donors. Incidence of aneuploidy and structural abnormality were 0.8%(hypohaploidy)and 2.81%, respectively. This result suggest that the hamster serum may be more suitable to the further maturity of hamster ova in vitro and contribute to the growth and development of male pronuclear chromosome.
