Abstract
The callus of Eucommia ulmoides was used to study the effect of inoculum amount, culture time and times of subculture on callus growth and secondary metabolites production. The results revealed that the callus growth of subculture was suitable when inoculum amount was 0.35 g. The callus growth curve was as S. The biggest increment of calli was on 20 d after subculture. The biggest content of chlorogenic acid and flavonoids was in 16 d culture. In the subculture course of callus, the shoot and leaf callus increment, chlorognic acid and flavonoids contents reach to peak in the fourth times. The cotyledon callus increment and flavonoids contents reach to peak in the fifth times, chlorogenic acid contents was in the fourth times. Chlorogenic acid contents was highest in callus of leaf and flavonoids contents was highest in cullus of hypocotyl in the callus of different sources.
