Abstract
Objective To observe the effect of sitagliptin on metabolic parameters ,serum visfatin level on high-fat-fed obesity rats. Methods Forty Sprague Dawley rats were randomly assigned into normal diet group (ND)(n=10)and high fat diet group(HD)(n=30). 16 weeks later,obese models(n=16)were established from HD group and were randomly assigned into obesity group (OB)(n=8)and sita-gliptin group(SITA)(n=8).Body weight(BW),fasting blood glucose(FBG),triglyceride(TG),total cholesterol(TCH),high-density lipo-protein(HDL-C),low-density lipoprotein(LDL-CC),fasting insulin(FINS),serum visfatin were measured before and after 6 weeks of treat-ment.Calculate HOMA-IR.Results The levels of BW,FBG,TG,TCH,LDL-C,FINS,HOMA-IR,serum visfatin in HD group were higher while the level of HDL-C was lower than those of ND group with a statistical significance (P<0.05).After treatment with sitagliptin,the lev-els of BW,FBG,TG,TCH,LDL-C,FINS,HOMA-IR,serum visfatin of SITA group were lower while the level of HDL-C was higher than those of OB group with a statistical significance (P<0.05).Pearson correlation analysis showed that serum visfatin was positively correlated with BW,FGB,FINS,HOMA-IR,TCH,LDL-C and negative correlated with HDL-C.Conclusion Obesity rat models induced by high-fat-fed show glucose and lipid metabolic disorders and insulin resistance .Serum visfatin levels are evaluated in obese rats , and sitagliptin intervention could lower serum visfatin levels in obese rats and improve the glucose and lipid metabolism and reduce insulin resistance .
