Effects of different culture factors on callus rate of eggplant anthers.

Abstract

The anthers of Xi'an eggplant variety were cultured under different hormones,carbon sources,amino acids and AgNO3to study the effects on the callus rate for establishing the optimum culture condition.The results showed that: 1) the callus rate was nearly zero when a single hormone was used in the medium,total callus rate and callus rate of central anther was above 18.0% and 4.0% when auxin and cytokinin was used in the medium simultaneously and the optimum medium was 0.2 mg/L 2,4-D+1.0 mg/L NAA+0.25 mg/L KT.2) totall callus rate was 57.14% when 2% sucrose was used in the medium and the callus rate of central anther all was above 12% when 2% sucrose and 6.5% glucose was used in the medium seperatly.3)the total callus rate and callus rate of central anther of treatments with adding amino acids and AgNO3was higher than CK.The optimum medium for improving anther callus rate of Xi'an eggplant variety was MS+0.2 mg/L 2,4-D+1.0 mg/L NAA+0.25 mg/L KT+2% sucrose +100 mg/L Ser.
