Effects of CN-100 on the functions of leukocytes in carrageenin and zymosan-induced pleurisy of rat.

The effects of 2- (10, 11-dihydro-l0-oxodibenzo [b, f] thiepin-2-yl) propionic acid (CN-100), a novel nonsteroidal anti-inflammatory drug, on the functions of leukocytes in carrageenin and zymosan-induced pleurisy of rats were investigated. The intrapleural injection of carrageenin and zymosan produced an acute inflammatory reaction, exudate formation, leukocyte mobilization and protein exudation in pleural cavity, and they have more potent activity than that of carrageenin.The acid phosphatase activity and TBA reactive substance were found to increase markedly in the supernatant of pleural fluids of rat with carrageenin and zymosan-induced pleurisy. In this model, CN-100 given orally inhibited significantly the exudate formation, leukocyte mobilization, protein exudation, acid phosphatase activity and TBA reactive substance in pleural cavity, and its inhibitory effect was almost equipotent as those of indomethacin and pranoprofen. These finding indicated that CN-100 has an inhibitory activities on the leukocyte functions as potent as that of indomethacin in local inflammatory reactions.From these results, it is suggest that CN-100 is suitable for clinical application.
