Isolation and diversity study of Actinomycetes associated with Marine Sponge Hymeniacidon perleve

Abstract

We isolated 244 strains culturable symbiotic actinomycetes from marine sponges Hymeniacidon perleve using 13 kinds of media. Among them 16S rDNA sequences of 106 typical strains were amplified. We analyzed RFLP type of the actinomycetes by digestion of 16S rDNA fragments with HhaI, and they showed 11 different types. Also the RFLP was analyisised with Vector NTI, the two results are similar. Using HhaI RFLP type, we could identify the actinomycete at the genus level. Phylogentic analysis using 16S rDNA sequences showed that most of actinomycetes belonged to Streptomycetes. The other strains belonged to Nocardiopsis, Pseudonocardia, Nocardia, Micromonospora, Rhodococcus, Actinoalloteichus. There are three candidates for new species: strain 124, 177 and B43.
