A Study on the Purification of Total Alkaloids from Xiatianwu Alkalods

Abstract

Objective To study the methods and technological parameters of the purification process of total alkaloids from herb using cation exchange resin.Methods In order to select the optimal technological parameters,the content of total alkaloids was determined by spectrophotometry.The content of protopine and tetrahydropalmatine was determined by high performance liquid chromatography.Results One gram cation exchange resin could absorb 31.33 mg/g of the total alkaloids.The optimized condition for purification was as follows: the concentration of the sample solution was 1 g/ml,and the flow rate was 1 ml/min.80% alcohol was used as the eluent and the quantity of the eluent was about three times the bed volume 89.5% of the total alkaloids could be washed off.The total alkaloids exceeded 70% after acid-alkali precipitation.Conclusion Cation exchange resin can well absorb the total alkaloids from herb.It can be used to pureify total alkaloids from Corydalis Decumbentis Rhizoma.
