Purification of total alkaloids from Corydalis Rhizoma based on evaluating liveness of extracorporeal anti-ulcer about cell "GSE-1"

Abstract

AIM To optimize and screen the purity of total alkaloids from Corydalis Rhizoma with the aid of GSE-1.METHODS The purification of the total alkaloids was performed with organic solution and on macroporous adsorptive resins.Acid dye colorimetry was used to determine total alkaloids from R.Corydalis.MTT assay was used to evaluate the effect of depurantive total alkaloids on GSE-1 proliferation,and the correlation between the purity and potency of total alkaloids was studied.RESULTS The optimal extraction was that the purified extract was dissolved with 1 mol/L hydrochloric acid.And then the impurities were removed by double volume of chloroform,ammonified,chloroform-acetoacetate-n-butanol extraction,three layers of organic solvent were collected and combined.CONCLUSION This process is reasonable and feasible the obtained purity of the total alkaloids is more than 90%,with a explicit connection resistance to ulcer of GSE-1 in vitro.
