Abstract
Objective Using anti CD3 monoconal antibody to substitute cyclosporin A for the establishment of the human B lymphoblastoid cell line. Methods The lymphocyte were separated from the peripheral blood. Then the anti CD3 monoconal antibody and the cyclosporin A were used as immunosuppressive agent to eatablish human B lymphoblastoid cell line. Results After 4 week culture the percentage of CD3+cells in anti CD3 monoconal antibody group was decreased from 59 65%to 52 78%, but was increased to 61.15%(P 0.05)in cyclosporin A group. On the other hand, the percentages of CD19+cells in anti CD3 monoconal antibody group and cyclosporine A group were increased from 5.39%to 12.70%and 8.47%respectively. The difference between these two groups was significant(P 0.01). Conclusion The anti CD3 monoconal antibody has the same enhancive effect as cyclosporin A on the establishment of B lymphocyte like cell line and can protect the B lymphocyte from regression in vitro.
