Abstract
Objective:To study the synergistic effect of combined use of cisplatin,docetaxel and hyperthermia on human lung adenocarcinoma A549 cell in vitro.Method:A549 cell was treated with different drugs,temperatures.Cell viability and growth were measured by means of MTT assay.The effect of combined use of cisplatin,docetaxel and hyperthermia were examined by means of morphologic observation and flow cytometry (FCM).Results:The significantly proliferation inhibition effect were observed when using either hyperthermia or drugs alone(P0.05),with a temperature or dose-dependent manner.Hyperthermia enhanced the proliferation inhibition rates compare with drug alone(P0.01).Significant synergistic effect was observed when cisplatin combined with docetaxel.The apoptosis ratio of combined use of cisplatin 2μg/ml,docetaxel 1μg/ml at 42℃ was higher than other groups.The results were coincident with morphologic observation.Conclusion:Synergistic antitumor effects of combined use of cisplatin,docetaxel and hyperthermia on A549 were prior to other groups,including significantly proliferation inhibition and apoptosis induction.
